For the most part, people do inseminations the day after a positive OPK, in other words, the day of ovulation. Some centers do two IUI, which would be the day of a positive OPK and the day after as just stated.

If you're doing the IntraCervical Insemination at home, then either do it the night of the Positive OPK or the morning after :)
